Abstract:
A sensitive and accurate method was designed by chemists from University of Mazandaran, Northern Iran, to measure antioxidant compounds, including vitamin C, in biological samples.

In addition to accurately measuring vitamin C in drug products, this method can also be used in measuring vitamin C at concentrations of 0.01 µg/mL in standard samples.

Taking into account the importance of the daily amount of vitamin C consumed by human body as food supplement, the researchers tried to introduce a selective, sensitive and accurate method to measure vitamin C in medical products based on its antioxidant properties.

In this research, the increase in sensitivity and decrease in diagnose limit of chemiluminescence system was made possible by producing gold/silver nanoalloy as the catalyst and introducing it in combination with Amplex red. Amplex red has unique luminescence properties (its oxidized form has fluorescence - chemiluminescence ability), and it is used as the probe. The observation of fluorescence caused by Amplex red in the absence of vitamin C a few minutes after the end of luminescence reaction confirms the presence of hydrogen peroxide as the oxidant in chemiluminescence system. In fact, hydrogen peroxide and its other active forms make possible the oxidation of Amplex red and its conversion into fluorescence form. Vitamin C, having antioxidant properties, is able to decompose hydrogen peroxide and disturb chemiluminescence reaction. Therefore, the designed luminescence system was used in the measurement of vitamin C in medical products.

Among the characteristics of the research, mention can be made of the simultaneous use of gold/silver alloy and Amplex red compound as strengthening agents of peroxyoxalate chemiluminescence system, and dual performance of Amplex red as fluorescence agent and probe that expresses the mechanism of vitamin C effect on chemiluminescence system.

Results of the research have been published in Journal of Luminescence, vol. 134, issue 3, February 2013, pp. 195-200.
