Abstract:
A novel single-walled carbon nanotubes(SWNTs)-molecular beacons(MBs) -daunoru -bicin(DNR) conjugate was devised as a targeted sensing system to detect S. typhimurium. The system, signified as SWNTs-MB(DNR), was fabricated by covalently coupling of a MB included complementary sequence targeted to the SSeC of the bacteria through EDC/Sulfo-NHS chemistry and insetting daunorubicin to the stem region of the molecular beacon on carbon nanotubes before adding the target sequence.

The fluorescence of daunorubicin, originally, was fairly week because of dual fluorescence quenching. When mixing target sequence with SWNTs-MBs(DNR), it was seen that the daunorubicin was discharged from hairpin stem due to the formation of rigid structure between the loop of the MB and the target sequence, which resulted in increase of fluorescence intensity substantially and attained to target quantitation. On the contrary, mismatch oligonucleotides of other Salmonella sps. with different sero types did not enhance the fluorescence intensity considerably, which represented a high selectivity and specificity of the biosensor. The experimental results demonstrated patently that the recovery of fluorescence of daunorubicin is proportion to the concentration of the target DNA with the range 0.2-0.7ìM and the low detection limit is 50nM. Consequently, the SWNTs-MBs(DNR) architecture can be an excellent platform to take place of the common molecular beacon easily and has the potential to study MB based on aptamer binding to the SWNTs.
